The ability of uncouplers to poke holes in the inner mitochondrial membrane would reduce the amount of ATP produced. This is because protons could move in a free direction through the membrane instead of powering the synthesis of ATP. For example, as an uncoupler, dinitrophenol can separate the flow of electrons and the pumping of hydrogen protons for the synthesis of ATP. The reduction in proton concentration leads to the blockage of the ATP formation process.
