The sample was obtained from the Californian population of pine. As a source of pathogenic fungi, a single wild-type perithecium was taken. It is a fruiting body of fungi. The perithecium was removed on the Petri dish with water agar in a drop of water in sterile conditions. After two days of cultivation at room temperature, three hyphal tips were aseptically transformed on potato dextrose agar. According to Sidhu, this cultural media is common for fungi cultivation. For F1, F2, and F3 generation obtaining, the authors collected ten ascospores from each perithecium. Totally, 30 progenies were obtained for each generation. For the experiment with the generation of fungi with high and low levels of virulence crossing, 100 progenies were taken from one perithecium.
